This time-lapse movie shows GFP-tagged ROD, a protein found at the condensed chromosome kinetochore, during the waves of synchronous mitoses. The sequence shows 1. kinetochore alignment at the cell equator in metaphase, 2. the splitting of sister chromosomes at anaphase onset and 3. kinetochore movement during anaphase until they reach the spindle poles in telophase. When the chromosomes decondense, the GFP-ROD signal disappears.
Time lapse 4D spinning disk confocal microscopy images were acquired every 8 seconds and are played at 6 frames/second. The images were acquired with a Nikon microscope (a X40 objective) and a Perkin-Elmer laser spinning disk. The program used to capture, pseudocolor, and process the images was Metamorph.
