Images were acquired with the Leica TCS AOBS SP2 confocal microscopy using a 63x oil immersion objective (NA 1.40) and an argon/krypton ion laser at wavelengths of 488 nm and an helium/neon 543 nm laser. The fluorescence signals were detected at 500-541 nm and 604-674 nm respectively. Z-series profiles of 30 optical sections were collected at intervals of 1.42 micrometers with a line average of 4 to reduce noise. Maximum projection of the series was applied. After confocal acquisition, z-stack was analysed with IMARIS software to adjust colours and to create 3D renders of chloroplasts.
