Description

The contractile vacuole, an organelle used by many protists for osmoregulation of water and some ions, such as calcium, is connected to the cell surface in Tetrahymena usually by two “pores," indentations of the plasma membrane. A spent food vacuole lies nearby. TEM taken on 1/8/74 by R. Wolf with Hitachi HU11A operating at 75kV. Neg. 17,500X. The raw negative was scanned with an Epson Perfection V750 Pro and this high resolution image is best used for quantitative analysis. Technical Details

Standard glutaraldehyde fixation followed by osmium tetroxide, dehydrated in alcohol and embedded in an epoxy resin. Microtome sections prepared at approximately 75nm thickness. Additional information available at (http://www5.pbrc.hawaii.edu/allen/).
