EM images of chromatin fibers reconstituted from tandem arrays of a strong nucleosome positioning sequence flanked by 'linker' DNA, creating fibers containing 22 nucleosomes and a nucleosome repeat length (NRL) of 177 bp. Reconstitutes had saturating amounts of purified core histones and linker histones. (A) shows a montage of 'end on' views of fibers obtained by cryo-fixation and recorded in the frozen hydrated state.
