Cells of the unicellular photosynthetic alga Chlamydomonas reinhardtii were fixed, sectioned, and examined by transmission electron microscopy. In this section of the ac-5 mutant grown phototropically, stacks (S) are primarily composed of two thylakoids. See Fig 10 in Goodenough and Staehelin 1971 J Cell Biol 48:595-619.
Cells were fixed with glutaraldehyde, post fixed with osmium tetroxide, dehydrated, embedded in epoxy resin, and thin sections cut, stained and examined with a Hitachi HU-11C TEM.
