Secondary lysosomes contain a fairly dense matrix in well fixed cells. The content of these lysosomes seems to be released as a package into the phagoacidosome when they fuse to form the phagolysosome. Secondary lysosomes contain both hydrolases and substrate. The appearance of HRP shows that some substrates have access to these secondary lysosomes. TEM taken on 4/19/79 by R. Allen with Hitachi HU11A operating at 75kV. Neg. 10,000X. Bar = 0.5µm.
Standard glutaraldehyde fixation followed by osmium tetroxide, dehydrated in alcohol and embedded in an epoxy resin. Microtome sections prepared at approximately 75nm thickness. The negative was printed to paper and the image was scanned to Photoshop. This digitized image is available for qualitative analysis.
