Immunogold label specific for L1 antigen on DV-II. Monoclonal antibody to antigen L1 specifically labels DV-II and acidosomes but not lysosomes. Lysosomes are osmotically active so they tend to shrink when the cell is exposed to hyperosmotic fixative as occurred here. This vacuole was 5-8 min. old and contained 0.8µm latex beads. TEM taken on 3/26/92 by R. Allen with Zeiss 10A operating at 80kV. Neg. 19,800X. Bar = 0.2µm.
Glutaraldehyde fixation followed dehydration in alcohol and embedded in an epoxy resin. Microtome sections prepared at approximately 75nm thickness. The negative was printed to paper and the image was scanned to Photoshop. This image is suitable for qualitative analysis.
