To see if acidosomes contain hydrolases (digestive enzymes) we incubated cells in Gomori’s medium which in the presence of acid phosphatase and lead will form an electron-opaque reaction product. Acidosomes surrounding a DV-I are free of reaction product even though a nearby DV, presumably a DV-III, contains abundant reaction product. A few vesicles assumed to be lysosomes also contain some reaction product. This experiment demonstrated that acidosomes are not lysosomes and do not contain digestive enzymes at least not AcPase. Glutaraldehyde fixation followed by Gomori acid phosphatase treatment and dehydration in alcohol and embedding in an epoxy resin. Microtome sections prepared at approximately 75nm thickness. The negative was printed to paper and the image was scanned to Photoshop. This digitized image is available for qualitative analysis.
