Description

Cultured HeLa cells were arrested in metaphase using nocodazole, immunostained for Plk1 and Myc, and counter stained with DAPI. The montage illustrates the effect of restoring Bub1 or Bub1-T609A to Bub1 depleted cells. Images shown are maximum Z projections of convolved widefield images.

Technical Details

Metaphase arrested cells were formaldehyde fixed, permeabilized with Triton X-100, immunostained, and z-stack images obtained at 0.2 micron intervals with a Zeiss Axiovert 200M microscope and 63x 1.4 NA objective lens.
