A time-lapse movie showing the dorsal pouch formation and migration of heart-anchoring cells (HANCs) in the Drosophila embryo expressing ubi-DE-cad-GFP (Shg-GFP). Shg-positive HANCs corresponding to the leading edge of the dorsal pouch fold inside from the head epidermis at the time of dorsal pouch invagination. They migrate posteriorly toward the heart and, in stage 16, overlap the anterior aorta. Notice a triangular shape of the migrating HANCs cluster. Embryos expressing Shg-GFP were dechorionated in bleach (2 min), rinsed in water, placed on a coverslip in the appropriate orientation, and then mounted in Voltalef 3M oil. The embryos were visualized using a Zeiss LSM 510 Meta confocal microscope. Images compiled from 1.5-mm-spaced optical sections were collected every 6 min during 2 h. Time-lapse series were assembled and analyzed by using Volocity (Improvision) software.
