A 3D reconstruction of the cardiac outflow tract (OFT)(horizontal rotation) from a stage-16 wild-type embryo stained for Ladybird early (Lbe; blue), Tinman (Tin; red), and the DE-Cadherin Shotgun (Shg; white). Heart-anchoring cells (HANCs) (in blue) dorsally overlap the aorta. Staining with Tin reveals its expression in the heart and in the cephalic vascular rudiment cells (cvr). Shg is expressed by the cardioblasts, the HANCs, and the ring gland cells. Stage 16 embryos were stained with primary antibodies: mouse anti-Lbe, rabbit anti-Tim, rat anti-DE-cadherin. Species-specific secondary antibodies were conjugated to Alexa Fluor 488, Cy3 or Cy5. The triple stained embryos were visualized using a Zeiss LSM 510 Meta confocal microscope. Images representing 0.5 mm optical sections were compiled and processed by Volocity software (Improvision).
